10.2 APPENDIX B: SOLUTIONS COMPOSITION

Bopp's acid : 3.9ml of 0.4 mol/L HCl, 250ml of 0.4M KCl. Adjust to pH =2.2 by adding 1M KOH

LB ampicillin plates with IPTG and X-gal: 40µl of 100mM isopropyl-beta-D-thiogalactopyranoside ( IPTG) and 40µl of 40mg/ml 5-Bromo-4-Chloro-3-Indolyl-ß-D-galactopyranoside (X-gal) on the surface of a Luria-Bertrani ( LB) agar plate containing ampicillin (100µg/ml).

IPTG (100mM): 238mg of IPTG in 10ml deionised water. Filter-sterilise and store in 1ml aliquots at -20.

X-GAL (40mM): 400mg in 10ml dimethylformamide (prepare in fume cabinet, dimethylformamide is poisonous). Protect from light. Store at -20 oC.
